RESUMO

Interest in antiviral plant species has grown exponentially and some have been reported to have anti-HIV properties. This research aims to perform the bio-guided phytochemical fractionation by antiretroviral activity of Lafoensia pacari stem barks. This in vitro experimental study involved the preparation of plant material, obtention of ethanolic extract, fractionation, purification, identification and quantification of fractions, acid-base extraction, nuclear magnetic resonance, HIV-1 RT inhibition test and molecular docking studies. From the bio-guided fractionation by the antiretroviral activity there was a higher activity in the acetanolic subfractions, highlighting the acetate subfraction - neutrals with 60.98% of RT inhibition and ellagic acid with 88.61% of RT inhibition and absence of cytotoxicity. The macrophage lineage cytotoxicity assay showed that the chloroform fraction was more toxic than the acetate fraction. The analysis of the J-resolved spectrum in the aromatic region showed a singlet at 7.48 and 6.93 ppm which was identified as ellagic acid and gallic acid, respectively. The 5TIQ enzyme obtained better affinity parameter with the ellagic acid ligand, which was confirmed by the HSQC-1H-13C spectra. Gallic acid was also favorable to form interaction with the 5TIQ enzyme, being confirmed through the HSQC-1H-13C spectrum. From the PreADMET evaluation it was found that ellagic acid is a promising molecule for its RT inhibition activity and pharmacokinetic and toxicity parameters.

Bone defects are a global health problem. The Rigenera system allows the extraction of micro grafts rich in mesenchymal stem cells (MSCs). The objective of this study was to evaluate the bone regeneration process by grafts obtained by the Rigenera system in defects in the rats calvarian. Eighteen male Wistar rats were used, weighing 285 ± 29g, distributed in three groups (n = 6), where each animal was treatment and control, called G15-Control and G15-Treated (15 days); G30-Control and G30-Treated (30 days) and G60-Control and G60-Treated (60 days). Two 5mm diameter lesions were performed on each calvaria side. In the treated groups, autologous micrograft from xiphoid cartilage, obtained by the Rigenera system, were used. The other defect served as a control in all animals. The animals were euthanized at 15, 30 and 60 days after the surgery and the samples were processed for histochemistry. In the control groups, no signs of bone regeneration were observed, while in the treatment groups, areas of bone formation and activated mesenchymal tissue were verified. The Rigenera system was efficient in obtaining autologous micrograft for cell therapy in a critical calvaria defect in rats. Rigenera system can be widely used in the treatment of bone injuries.(AU)

The goal of this study was to determine the effect of stanozolol (ST) on kidney and liver of Wistar rats. Thirty-two male animals were divided into the following four groups: control group (CG); Control group-exercise (GCE); Group-steroid treatment (GT); Group treatment-steroid-exercise (GTE). Swimming was defined as exercise. The animals GT and GTE was submitted to subcutaneous injections, five days/week for 30 days, at a concentration of 5mg/kg ST diluted in 1mL/kg of sesame oil. The results showed an increase in weight gain in all animals submitted to ST and exercise from the 3rd week of use and increase in urinary excretion from the 5th week and the other urinalysis parameters were similar. The ST associated or not with physical activity reduced VEGF-A expression in the kidneys and hepatic glycogen, as well as histopathological changes in these organs. Regarding hematology, there was a decrease in leukocytes in the GTE. As for lymphocytes there was an increase in GT and a decrease in GTE, and in relation to the number of platelets, there was a decrease in GTE. In conclusion, the administration of stanozolol at 5.0mg/kg caused a structural change of kidney and liver in treated animals.(AU)

RESUMO

Many threats exist to reptile populations, environmental pollutants being one of them. Lizards and other reptiles are usually not taken into consideration in environmental risk assessments, with the use of surrogate species for their estimates. Unfortunately, not all pesticides have the same effects in the reptile species and on these surrogates, birds and mammals, some being more toxic in lizards. This difference brings the need to evaluate their toxicity in lizards to safeguard its protection. Studies in the last decades involving contaminants' toxicity in lizard species have increased, thus we proposed to gather these information in this comprehensive review. Through searches in databases about the toxicity of pesticides in lizards, 16 scientific articles were found. Most studies investigated locomotor performance, histopathology, oxidative stress, neurotoxicology, and genetic damage from diverse pesticides with different modes of action. Progress has been made to acquire data on lizard ecotoxicology and more research is needed to cover more variables, such as studies in the embryologic stage and different pesticides.

RESUMO

2,4-D is a selective pre- and postemergence herbicide used for several crops. It is hazardous for the environment and risk for humans; therefore, several studies attempt to evaluate its effects and consequences of its use. The nervous system is supposedly a target for this herbicide, and this comprehensive review gathers the information about animal models that have been used for the study of the neurotoxicity of 2,4-D. The studies used several methods to evaluate the neurotoxicity of this herbicide, most of which used rodents, mainly rats, two used fish, and one used chicken eggs. The main behavioral effect observed concerned alterations in locomotor patterns and reduced motor activity. Biochemical analysis showed decreased levels of serotonin (5-HT) and increased levels of its metabolites and increased or decreased levels of DA and its metabolites depending on the brain area analyzed. Hypomyelination is also a possible effect of 2,4-D when the exposure occurs during the proliferation and development of the oligodendrocytes. The worst neuropathologic effects were observed in fish. Since most studies focused on the neurotoxicity of 2,4-D in rodents, the effect it may have on other species and groups of animals, especially with different physiology, is unclear and it should be researched.

RESUMO

The aim of this systematic review was to determine whether bromelain is an effective drug for the control of pain and inflammation associated with third molar surgery. Randomized, controlled clinical trials on the subject were identified through a systematic search of the literature using the PubMed/MEDLINE, Scopus, and Cochrane Library databases. This review was conducted in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA). Two independent, blinded reviewers selected studies based on the inclusion criteria. Divergences of opinion were resolved by consensus. A meta-analysis was performed for the outcomes pain and trismus and was based on the inverse variance method for continuous outcomes, considering the mean difference (MD) and corresponding 95% confidence interval (CI). A total of 186 articles were initially retrieved from the databases. After the different stages of the selection process, five articles reporting data for a total 252 patients remained and were included in the review. Bromelain proved to be effective at controlling postoperative pain at 48-72h after surgery (P=0.03; MD -0.89, 95% CI -1.70 to -0.09), but did not achieve a significant effect in comparison to the control group with regard to oedema or trismus.

RESUMO

A new species of Astyanax is described from the upper Rio Paraguai basin, Mato Grosso State, Brazil. The new species can be distinguished from congeners by having the body intensely yellowish in life (v. silvery, reddish or lightly yellow) and by morphometric and meristics traits. Astyanax dolinae n. sp. cannot be assigned to any of the Astyanax species complex currently recognized for the genus. It is only known from the Dolina Água Milagrosa, a karstic sinkhole lake, entirely fed by groundwater, surrounded by Cerrado, the savannah-like vegetation of central South America.

RESUMO

AIM: The goal of this study was to increase knowledge about the antimicrobial activity of some synthetic Riparin-derived compounds, alone or in combination with fluoroquinolone antibiotics, against a strain of Staphylococcus aureus resistant to fluoroquinolone by way of overexpression of the NorA efflux pump. METHODS AND RESULTS: Microdilution tests showed that Riparins A and B did not show any significant antibacterial activity against Staph. aureus strains. On the other hand, the intrinsic antibacterial activity increased with increasing lipophilicity of the compounds, in the following order: Riparin-D (MIC 256 µg ml-1 ; Log P 2·95) < Riparin-C (MIC 102 µg ml-1 ; Log P 3·22) < Riparin-E (MIC 16 µg ml-1 ; Log P 3·57). The addition of all riparins to growth media at subinhibitory concentrations caused an increase in the antibacterial activity of antibiotics against the NorA-overexpressing test strain. Riparin-B, which has two methoxyl groups at the phenethyl moiety, showed the best modulatory effect. CONCLUSIONS: Riparin-E is a good anti-staphylococci agent, while Riparin-B functions as a NorA efflux pump inhibitor. SIGNIFICANCE AND IMPACT OF THE STUDY: Our data suggest the possibility of using Riparin-B in combination with norfloxacin or ciprofloxacin for therapy of infections caused by multi-drug resistant Staph. aureus.

No characteristic of living beings is as primal as breathing, and the lungs are the main organs in the respiratory system. This study aims to describe the macroscopic aspects of the trachea, bronchus and lung lobes and microscopic aspects of the bronchi of raccoon lungs and compare with data from the literature studies performed with wild and domestic mammals. We used three samples of Procyon cancrivorus, which were fixed in aqueous 10% formaldehyde, lungs and trachea were dissected and photographed with a digital camera (Sony a200 Camera, 10.2mpx). For the identification of microscopic characteristics, fragments were collected from each bronchus following routine histological techniques. The Procyon cancrivorus lung is divided into four lobes, with two right and left lobes. The trachea has about 31-34 rings. The extrapulmonary bronchi divides into left and right, where the right is divided into lobar bronchi cranial, middle, accessory and caudal lobes and the left in cranial and caudal, with their respective segmental bronchi. Microscopically the bronchial epithelium has prismatic pseudo-ciliated and goblet cells with bundles of smooth muscle fibers, plates of hyaline cartilage and elastic fibers. Knowledge of the morphology of these organs in wild species helps us in descriptive studies and / or comparisons between species.

RESUMO

A new species of the catfish genus Centromochlus (Auchenipteridae, Centromochlinae) is described. The new species is diagnosed by having numerous dark rounded blotches over the body and fins, dorsal-fin spine with serrations anteriorly and smooth posteriorly, anal fin of mature males with three unbranched and seven branched rays, anterior nuchal plate absent and posterior nuchal plate not extended ventrally. The new species is described from a small stream in the Estação Ecológica Serra Geral de Tocantins, a natural reserve in the centre of the Brazilian Cerrado, close to the watershed between the Rio Tocantins and the Rio São Francisco basins. The new species is possibly the sister taxon to the recently described Centromochlus meridionalis from the upper Rio Tapajós. Those two species share with Centromochlus perugiae, from the upper Amazon and upper Paraguay, derived features associated with the modified anal fin in sexually mature males.

RESUMO

Genetic variability at 11 microsatellite markers was analyzed in five naturalized/local Brazilian horse breeds or genetic groups. Blood samples were collected from 328 animals of the breeds Campeira (Santa Catarina State), Lavradeira (Roraima State), Pantaneira (Pantanal Mato-Grossense), Mangalarga Marchador (Minas Gerais State), as well as the genetic group Baixadeiro (Maranhão State), and the exotic breeds English Thoroughbred and Arab. We found significant genetic variability within evaluated microsatellite loci, with observed heterozygosis varying between 0.426 and 0.768 and polymorphism information content values of 0.751 to 0.914. All breeds showed high inbreeding coefficients and were not in Hardy-Weinberg equilibrium. The smallest genetic distance was seen between the Pantaneira and Arab breeds. The principal component analyzes and Bayesian approach demonstrated that the exotic breeds have had a significant influence on the genetic formation of the local breeds, with introgression of English Throroughbred in Pantaneira and Lavradeira, as well as genetic proximity between the Arab, Pantaneira and Mangalarga Marchador populations. This study shows the need to conserve traits acquired by naturalized horse breeds over centuries of natural selection in Brazil due to the genetic uniqueness of each group, suggesting a reduced gene flow between them. These results reinforce the need to include these herds in animal genetic resource conservation programs to maximize the genetic variability and conserve useful allele combinations.

RESUMO

Accumulating evidence indicates that genetic background influences the outcome of sepsis, which despite medical advances continues to be a major cause of morbidity and mortality. This study aimed to evaluate the influence of SNPs LTA +252A>G, TNF-863C>A and TNF-308G>A on susceptibility to sepsis, acute respiratory distress syndrome (ARDS), septic shock and sepsis mortality. A prospective case-control study was carried out in a Brazilian pediatric intensive care unit and included 490 septic pediatric patients submitted to mechanical ventilation and 610 healthy children. No SNP association was found with respect to sepsis susceptibility. Nevertheless, a haplotype was identified that was protective against sepsis (+252A/-863A/-308G; OR=0.65; p=0.03). We further observed protection against ARDS in TNF-308 GA genotype carriers (OR=0.29; p=0.0006) and -308A allele carriers (OR=0.40; p=0.003). In addition, increased risk for ARDS was detectable with the TNF-863 CA genotype (OR=1.83; p=0.01) and the -863A carrier status (OR=1.82; p=0.01). After stratification according to age, this outcome remained significantly associated with the -308GA genotype in infants. Finally, protection against sepsis-associated mortality was found for the TNF-308 GA genotype (OR=0.22; p=0.04). Overall, our findings document a protective effect of the TNF-308 GA genotype for the ARDS and sepsis mortality outcomes, further providing evidence for an increased risk of ARDS associated with the TNF-863 CA genotype. Trial registration (www.clinicaltrials.gov): NCT00792883.

RESUMO

Embryos of Caiman yacare were collected and subjected to the bone clearing and staining protocol in orderto analyze the ontogenetic patterns of ossification of the pectoral girdle and forelimb skeleton. The osseousstructure of the girdle and forelimbs of C. yacare begins to ossify starting at 30 days of incubation, withthe presence of dye retention in the scapula, coracoids, humerus, radius and ulna bones. During embryonicdevelopment, the autopodio of C. yacare has four bones in the carpus, the radial, ulnar, pisiform and carpaldistal 4+5 bone. Their ossification begins at 39 days of incubation with the radial, followed by the ulnar, and at54 days, the pisiform and the distal carpal 4 + 5. Each mesopodio has 5 metacarpi and are present 15 phalanges, two in digits I and V, three in digits II and IV, and four in digit III (phalangeal formula 2:3:4:3:2). Ossificationof the metacarpi starts at 27 days of incubation, following the sequence MCII=MCIII=MCIV>MCI>MCV.The first phalanges begin the process of ossification on day 36, continuing up to the last day of incubation. The sequence of ossification of the proximal phalanges is PPI=PPII=PPIII>PPIV=PPV, that of the medialphalanges is MPII>MPpIII>MPdIII>MPIV, and that of the distal phalanges is DPI>DPII>DPIII>DPV>DPIV.The ontogenetic pattern of the bones of the forepaw of C. yacare generally differs from that of other reptiles, although there are some similarities.

RESUMO

The determination of embryonic stages is very useful in comparative and evolutionary anatomy, experimentalresearch and ecological studies. Although tables of development stages already exist for some crocodilianspecies, no table has yet been published for Melanosuchus niger. Therefore, the aim of this work is to monitorthe development of this species of Alligatoridae and characterize its stages based on the main morphologicaltransformations the embryo undergoes during the incubation process. Under natural conditions, two sampleswere collected daily in the Lake Cuniã Extractive Reserve in Rondônia, Brazil, making a total of 186 eggs. The incubation period was 90 days at an ambient temperature varying from 25.2 to 31.7 °C. We established24 embryonic stages between the formation of the pharyngeal arch and hatching. Primordia for the sensoryorgans and somites were observed in the early stages. The buds of the pelvic and thoracic limbs emergedsimultaneously, also in the early stages. An analysis of these structures showed evidence of considerableevolutionary modifications when compared with embryos of different species. In stage 12 we observed subtlepigmentation in the region of the head and, in stage 25, the pigmentation and striped pattern were typical ofthe newly hatched specimen. The eyetooth was visible in stage 18, disappearing within the first few weeks afterhatching. The vitellus was completely absorbed in the penultimate stage. Direct comparisons of the table ofembryonic stages were made between M. niger and other crocodilian taxa, and the main differences identifiedwere chronological variations.

RESUMO

Various essential oils are rich in carvacrol, a monoterpenic phenol isomeric with thymol. This study was undertaken to assess the vasorelaxant effects of thymol and carvacrol in rat isolated aorta and the putative mechanisms underlying these effects. Thymol and carvacrol produced a concentration-dependent relaxation on the aortic ring preparations pre-contracted using KCl (IC(50) value of 64.40 +/- 4.41 and 78.80 +/- 11.91 microm, respectively) or using phenylephrine (PHE, 0.1 microm) (IC(50) value of 106.40 +/- 11.37 and 145.40 +/- 6.07 microm, respectively) and inhibited the concentration-response curves of aortic rings to PHE or KCl. In Ca(2+)-free medium with ethylene glycol-bis(2-aminoethylether)-N,N,N',N'-tetraacetic acid (2 mm), thymol and carvacrol both at 1000 microm completely abolished the phasic component of PHE-induced endothelium-containing ring contractions. At 400 microm, thymol and carvacrol significantly reduced the CaCl(2)-induced contractions in Ca(2+)-free medium. Furthermore, both thymol and carvacrol (300 and 1000 microm) significantly reduced the contraction evoked by phorbol dibutyrate (1 microm), an activator of protein kinase C. Magnitude of this inhibitory effect was enhanced in the presence of the Ca2+ pump inhibitor, thapsigargin (1 microm). At 1000 microm, neither thymol nor carvacrol altered the resting potential of vascular smooth muscle cells. In conclusion, thymol and carvacrol induced an endothelium-independent relaxation in rat isolated aorta, an effect that seems mediated through some mechanisms probably involving a transduction pathway between Ca(2+) release from sarcoplasmic reticulum and/or regulation of the Ca2+ sensitivity of the contractile system. Moreover, it's conceivable that thymol and carvacrol, at low concentrations, block the Ca(2+) influx through the membrane.

RESUMO

Scleral ossicles are bony plates in the eye sockets of many vertebrates, whose morphology, development and position differ in distinct groups of vertebrates. To investigate the development, number, shape and arrangementof the scleral ossicles of Podocnemis expansa, 15 embryos were collected, starting from the 18th day of natural incubation and 16 adult specimens belonging to the LAPAS collection, whose eye sockets were removed. The embryos and eye sockets were subjected to bone diaphanization and staining techniques. The scleral ossicles in this species of Testudines were found to be quadrangular, varying in number from 10 to 13, with 11.25 ± 0.93 in the right eye and 11.43 ± 0.81 in the left eye, occupying a position close to the anterior edge of the sclera. The ossicles develop intramembranously and begin to present ossification centers at stage 21, with ten ossification centers in the right eye and eight in the left eye. In stage 23, all the ossicles of the sclerotic ring increase in length and approach the adjacent ossicles, forming a ring. In stage 26, the scleral ossiclesare already arranged in their definitive form.

RESUMO

An outbreak of coccidioidomycosis is described that involved three individuals and eight of their dogs, who had engaged in a successful hunt for nine-banded armadillos (Dasypus novemcinctus) in the environs of Oeiras, a community in Brazil's north eastern state of Piauí. Diagnosis was based on clinical, serological and cultural findings. Four of 24 soil samples collected in and around the burrow of an armadillo yielded cultures of Coccidioides immitis, thus establishing the endemicity of that mould in the state of Piauí. A literature review revealed that C. immitis, aside from that state, is endemic in three other Brazilian states--Bahia, Ceará and Maranhão. These four contiguous states have semi-arid regions where climatic conditions and their flora are similar to those that exist in C. immitis's endemic regions in North, Central and South America.
